Nghiên cứu tiến hành xây dựng và thẩm định phương pháp định lượng vitamin C trong viên nén sinh tồn bằng sắc ký lỏng hiệu năng cao (HPLC). Điều kiện sắc ký gồm cột C18 (250 x 4,6 mm, 5µm), pha động dung dịch acid acetic 0,1% – methanol (90 : 10, v/v), tốc độ dòng 0,5 mL/phút, thể tích tiêm 10 µL và bước sóng phát hiện tại 245 nm. Mẫu được xử lý bằng phương pháp chiết siêu âm một lần với dung dịch TFA 0,1% trong 15 phút. Phương pháp đạt yêu cầu thẩm định theo yêu cầu của ICH với tính tương thích hệ thống và độ đặc hiệu cao. Phương pháp có khoảng tuyến tính từ 0,25 – 500 µg/mL với hệ số tương quan R² = 0,9999; giới hạn phát hiện (LOD) và giới hạn định lượng (LOQ) lần lượt là 0,10 µg/mL và 0,25 µg/mL. Độ chính xác của phương pháp đạt yêu cầu với giá trị RSD trong ngày và khác ngày đều nhỏ hơn 2%, đồng thời độ thu hồi nằm trong khoảng 95 – 105% với RSD < 2%. Các kết quả khảo sát đạt yêu cầu về thẩm định, có thể áp dụng để xác định hàm lượng vitamin C trong viên nén sinh tồn.
