Study on extraction, isolation and purification of acid ursolic from coffee Arabica leaves for reference standard establishment

Abstract
Currently, ursolic acid is found in many medicinal herbs and has been proven to have pharmacological activities, including anti-oxidation, anti-inflammation, antibacterial, antiviral, anti-diabetic, and anti-cancer effects. In the Vietnamese Pharmacopoeia V, ursolic acid is being used as a standard substance in the qualitative determination of some medicinal herbs such as Ocimum tenuiflorum L., Cornus officinalis, Eriobotrya japonica. Ursolic acid is also considered a marker for medicinal herbs such as Nepeta stenantha, Guiera senegalensis. To contribute to the standardization of medicinal herbs containing ursolic acid, it is necessary to use a standard substance of its with high purity. Currently, there is no ursolic acid standard substance available in Vietnam at a reasonable price. In this study, 1.08 g of ursolic acid was extracted, isolated and purified from 1 kg of dried arabica coffee leaves. The purified product structure was confirmed by spectroscopic methods (IR, HRMS, NMR) through comparison with the reference standard while the content determined by HPLC-PDA method reached 95.4%.
