Establishing a reference standard of astragaloside IV isolated from Radix Astragalus

Abstract

In China and Vietnam, Radix Astragalus has been widely used as a traditional herbal. With reference to the quality of Radix Astragalus in Vietnam Pharmacopoeia V, Chinese Pharmacopoeia (CP 2020), its content of astragaloside IV quantified by HPLC/ELSD is specified to be not less than 0.04 %. For the quality control of Radix Astragalus, the establishment of astragaloside IV as an affordable-price reference standard is prerequisite in Vietnam. In this study, 3,5 g of astragaloside IV was isolated and purified from 2,5 kg of Radix Astragalus. The purified substance whose structure was confirmed astragaloside IV by 1H, 13C-NMR, HR-MS, IR. Astragaloside IV was packaged in a 10 mg standard tube, the content of astragaloside IV in our purified samples was determined to be of 96.0 % by HPLC-ELSD analysis.
